Cow's milk contains two major sources of protein, casein (80%) and whey (20%) . Like whey protein, casein is comprised of several protein fractions - alpha casein, beta casein, gamma casein, delta casein, and kappa casein - that although are structurally very similar, differ in their functional properties, amino acid composition and size. These individual casein proteins alone are not very soluble in the aqueous environment of milk. However, in whole milk, they aggregate together to form larger complexes - held together by calcium phosphate - that are porous and can be easily dispersed in the watery part of milk. These complexes - or structures - are called casein micelles, and are the native form of casein found in milk. Thus micellar casein is completely undenatured and highly digestable, and retains the full biological activity of all the casein fractions it contains. Micellar casein has been shown to have many health benefits; including immuno-modulatory effects, anti-bacterial action, and raised levels of HDL (good cholestrol).
Micellar casein, like whey, is rich in essential amino acids (50%). It's also high in glutamine (10%) and glycomacropeptides (GMP's). GMP's, whilst also found in whey protein derived from cheese manufacture (WPC), are found most abundantly in undenatured micellar casein (from the kappa casein fraction). These biologically active peptides produce a number of beneficial effects, including the release of cholecystokinin (CKK) - a hormone that helps suppress appetite.
The rate of absorption of dietary proteins largely determine their effects on protein synthesis and protein breakdown. Consuming casein results in a slow release of amino acids into plasma, which only moderately stimulates the rate of whole body protein synthesis by 31% (compared to 68% for whey). This slower absorption is caused by casein forming clots in the stomach when ingested, which in turn, slows down gastric emptying. Moreover, the bioactive peptides within casein also slow down gastrointestinal motility - further slowing down the release and subsequent absorption of amino acids. This results in a very different metabolic response when compared to faster proteins such as whey. Casein has been shown to inhibit muscle breakdown by 34% over a 7 hour period, with only a minimal increase in protein oxidation.
Therefore, micellar casein based formulations have obvious applications for those populations wishing to prevent muscle catabolism (wasting conditions, athletes etc), as well as being the ideal protein source for situations where it is not possible to practice repeat protein feedings i.e. during sleep. Milk Protein Concentrate (MPC) is the richest source of micellar casein, as the proteins found in MPC are in the exact same ratios as those found in bovine milk (80% micellar casein / 20%). MPC is produced through the same low temperature process of ultrafiltration as is whey protein concentrate (WPC) - albeit with skimmed milk is the starting material as opposed to sweet dairy whey.
