L Leucine Powder

L-Leucine, is one of the three amino acids referred to as branched-chain amino acids. Leucine comprises about eight percent of the total amino acid count in your body's protein structures and is the fourth most concentrated amino acid in skeletal muscle tissue.

Leucine is unique in its ability to stimulate skeletal muscle protein synthesis and has a far greater stimulatory effect on protein synthesis (10 fold ) than any other amino acid. Leucine is what's known as a "limiting nutrient" - meaning that you must have enough L-Leucine in proportion to other amino acids in order for your body to make use of what you eat. Supplementing with upwards of 2gms of L-leucine per meal has been shown avoid this rate limiting effect, which is more pronounced in older populations.

The main anabolic pathway through which leucine exerts its anabolic action is via mTOR (Mammalian Target of Rapamycin), one of the body's regulators of protein synthesis. By activating/inactivating specific proteins, leucine both triggers protein synthesis as well as increases the actual capacity of a cell to conduct protein synthesis (combining leucine with glutamine increases this upregulation of cellular capacity even further). ATP levels also play a role in activation of mTOR. It is activated when ATP levels are high, and blocked when ATP levels are low. Whilst supplements such as creatine and ribose directly elevate levels of ATP, leucine can mitigate this inhibition on protein synthesis caused through low ATP levels. Whilst leucine’s primary role appears to be increasing protein synthesis, it also decreases protein breakdown (anti-catabolism) through a variety of mechanisms, such as increasing HMB synthesis (a metabolite of leucine), sparing leucine levels in muscle, as well as increasing levels of insulin - which is a major regulator of protein degradation. Although leucine and insulin’s effects appear to be independent of each other, when both insulin and leucine levels increase simultaneously, they have a synergistic effect on protein synthesis. Leucine also enhances insulin's response to carbohydrates, which further maximises the body’s anabolic response.

With all of leucine's effects seemingly being strictly positive, it is worth noting that when administered alone in too high of a dose, it can lower the concentration of other amino acids, particular other BCAAs.
